After reading this chapter you should be able to:
  • Explain the basis of the molecular attraction that causes nucleic acid hybridization reactions to occur; explain the difference between the probe and target sequences in hybridization assays.
  • Describe what is meant by a 'primer pair' used in a PCR assay, and explain how the sequence of these primers determines the product amplified in the PCR process.
  • Describe the typical characteristics of restriction endonucleases with regard to their selection of cleavage sites on DNA and the types of fragment ends that are produced.
  • Describe the general steps used to perform a Southern blot and the type of information it provides.
  • Differentiate between a cDNA and a genomic clone.
  • Describe typical features of a bacterial plasmid used for cloning DNA.
