Mutations in tumor suppressor genes are recessive and, thus, mutations in both copies of the gene are usually required for transformation. As it is very difficult to identify the loss of function of a single gene in a cell, much of the initial information relating to tumor suppressor genes was obtained by studying a range of inherited cancer syndromes (Table 41.3).
