Affinity chromatography is a convenient and specific method for purification of proteins. A porous chromatography column matrix is derivatized with a ligand that interacts with, or binds to, a specific protein in a complex mixture. The protein of interest will be selectively and specifically bound to the ligand while the others wash through the column. The bound protein can then be eluted by a high salt concentration, mild denaturation, or by a soluble form of the ligand or ligand analogs (see Chapter 5).
